چکیده

It was in 1909 that Young, in the laboratory of Harden, isolated from fermenting yeast the fructose bisphosphate (the Harden and Young ester) that was later identified by Levene & Raymond (1928) as fructose 1,6-bisphosphate. Some 71 years elapsed until a second fructose bisphosphate was isolated from living cells and identified as fructose 2,6bisphosphate (Van Schaftingen et al., 1980c). In contrast with its elder brother, Fru-2,6-P2 is not an intermediary metabolite of glycolysis, but its most potent regulator. In the present review, we will summarize the work that led to its discovery and we will try to delineate its potential role in the control of metabolism in various organisms.

PFK-2/FBPase-2: maker and breaker of the essential biofactor fructose-2,6-bisphosphate.

Fructose-2,6-bisphosphate is responsible for mediating glucagon-stimulated gluconeogenesis in the liver. This discovery has led to the realization that this compound plays a significant role in directing carbohydrate fluxes in all eukaryotes. Biophysical studies of the enzyme that both synthesizes and degrades this biofactor have yielded insight into its molecular enzymology. Moreover, the meta...

Fructose 2,6-bisphosphate and 6-phosphofructo-2-kinase during liver regeneration.

Glycogen and fructose 2,6-bisphosphate levels in rat liver decreased quickly after partial hepatectomy. After 7 days the glycogen level was normalized and fructose 2,6-bisphosphate concentration still remained low. The 'active' (non-phosphorylated) form of 6-phosphofructo-2-kinase varied in parallel with fructose 2,6-bisphosphate levels, whereas the 'total' activity of the enzyme decreased only...

Increasing fructose 2,6-bisphosphate overcomes hepatic insulin resistance of type 2 diabetes.

Hepatic glucose production is increased as a metabolic consequence of insulin resistance in type 2 diabetes. Because fructose 2,6-bisphosphate is an important regulator of hepatic glucose production, we used adenovirus-mediated enzyme overexpression to increase hepatic fructose 2,6-bisphosphate to determine if the hyperglycemia in KK mice, polygenic models of type 2 diabetes, could be ameliorat...

Fructose 2,6-bisphosphate hydrolyzing enzymes in higher plants.

The phosphatases that hydrolyze fructose 2,6-bisphosphate in a crude spinach (Spinacia oleracea L.) leaf extract were separated by chromatography on blue Sepharose, into three fractions, referred to as phosphatases I, II, and III, which were further purified by various means. Phosphatase I hydrolyzed fructose 2,6-bisphosphate, with a K(m) value of 30 micromolar, to a mixture of fructose 2-phosp...

Effect of diabetes, insulin, starvation, and refeeding on the level of rat hepatic fructose 2,6-bisphosphate.

The influence of alloxan diabetes and starvation for 72 h on the level of rat hepatic fructose 2,6-biphosphate was investigated. Both diabetes and starvation decreased the level to 10% of the value found in livers of normal, fed rats (10 nmol/g liver). The activity of the enzyme responsible for the synthesis of fructose 2,6-bisphosphate, 6-phosphofructo 2-kinase, was also decreased in livers of...
